At the Drug Screening Platform (DSP), we provide expert support for Chemical Biology and Drug Discovery projects. Our platform accepts projects from the entire IRB Barcelona research community, as well as from external projects, both from academia and industry. Our expert team provides tailored experimental support for a wide range of targets and technologies during the Candidate Discovery phase, from High-Throughput Screening (HTS) to Lead Optimisation The use of state-of-the-art equipment (listed below) together with robust analysis software, allows you to run your projects following industrial standards in an accurate, fast and reliable manner.

Biostatistics / Bioinformatics

The mission of the Biostatistics / Bioinformatics Unit is to carry out collaborative research and offer consultation and quantitative research resources to all IRB Barcelona research groups. We help scientists by offering consulting services both in the short and long term in the following areas: Experimental design: sample size, study design, statistical methodology planning Data analysis: clinical or biomedical databases, genomics data Help with statistical methodology in own or other people's research Software: help in the use of software, software development to meet the special needs of data analysis or study design Methodological research is carried out and resources are developed in all the areas mentioned.

Advanced GENOMIC Technologies

The Functional Genomics Core Facility (FGCF) at the Institute for Research in Biomedicine (IRB Barcelona) offers comprehensive support for genomics and transcriptomics projects in tissues and cells. Our facility supports the entire IRB Barcelona research community, but also other scientists and groups in academia and industry. We offer tailor-made solutions for a wide range of projects, including those with very limited sample quantities, or samples with compromised quality. We also offer all interested researchers single-cell genomics, transcriptomics, and epigenomics solutions, in particular those requiring 10X Genomics microfluidics technology. The FGCF can support the following projects: Transcriptomics of tissues or cell cultures (mRNA-seq, miRNA-seq, sRNA-seq, and lncRNA-seq). Messenger RNA sequencing and single-cell multiomics using 10X Genomics technology. Whole genome sequencing (WGS). Chromatin immunoprecipitation and sequencing (ChIP-seq). RNA immunoprecipitation and sequencing (RIP-seq). High-throughput chromosomal conformation capture (Hi-C/3C-Seq). Amplicon sequencing and capture of regions of interest. Mouse Mutant

The facility was created in 2007. Our primary function is to generate genetically modified mouse models for users within IRB Barcelona. We are able to offer a comprehensive service for the generation of genetically altered mice including: Design of recombinant DNA molecules for genetic modification. Help with cloning protocols and reagents required to generate gene targeting and transgenic vectors. All cell-culture required for gene targeting in mouse embryonic stem (ES) cells, including expansion, transfection, drug selection, colony picking and colony expansion. Microinjection of DNA or ES cells into pre-implantation stage embryos. Electroporation of DNA/mRNA/proteins into pre-implantation stage embryos. Transfer of manipulated embryos into pseudo-pregnant foster mice. Subsequent processing of pups born to pseudopregnant fosters.

Mass spectrometry and proteomics

Modern proteomic tools are essential for understanding biological issues. The basic installation of mass spectrometry and proteomics (MSPCF) offers IRB Barcelona researchers and external users a series of techniques based on mass spectrometry (MS) and proteomics that can help improve the their knowledge of diseases, pathways, targets, and the effects of drugs. Develops and implements a wide range of methodologies based on mass spectrometry and applies them to various topics in structural biology, biochemistry, cell biology and, in clinics, in collaboration with IRB research groups and external researchers. . . Currently, the facility offers state-of-the-art applications in quantitative proteomics (differential expression protein analysis using targeted, multiplexing, and unlabeled strategies), high-performance post-translational modification (PTM) analysis, protein, structural MS ((biotinylation).-MS, crosslinking-MS, native MS; study of protein structure, protein complexes and their dynamic interactions) and high-throughput top-down proteomics (which characterizes intact proteins , i.e. proteoforms). The facility has extensive experience in proteomic analysis in all its approaches (e.g., intact proteins, classical proteomics, data interpretation, and analysis of large data sets) and provides knowledge about proteomics to the scientific community. Non-covalent protein-protein and protein-ligand interactions are also studied by MS and ion-coupled mobility in MS (IM / MS) in order to delve into the mechanisms of action of these proteins in biological processes.

Protein expression

The Protein Expression Core Facility was founded to offer high-performance expression cloning and screening (HTP) activities in which many variations of a protein (e.g., can be performed in parallel. Alternatively, HTP activities they can be directed to the genome of a particular organism (or family of organisms) to clone and call expression many different proteins from a single organism. In addition, the facility has the experience and equipment needed to produce and purify milligrams of protein from prokaryotic and eukaryotic expression hosts, currently E. coli, Sf9 (insect), and HEK293T or HEK293A (mammalian cells). Many of the protocols are automated, with the installation making full use of liquid handling robotics for HTP plate handling for small-scale expression screening (μg) and automated purification systems (Äkta Xpress) for larger-scale protein purification (mg). The facility also provides many high-quality reagents for protein cloning and expression, such as competent bacteriophage-resistant E.coli strains, specialized expression media, and recombinant enzymes. If necessary, the installation can also offer custom vector cloning and modification services.

ADM – Advanced digital microscopy

Advanced Digital Microscopy platform is a central unit of IRB Barcelona. It opened in January 2009 and offers 24/7 access and support to 12 state-of-the-art and conventional imaging instruments for life science research, from high content imaging to microscopy wide field, confocal/multiphoton and super resolution, to emerging techniques. for cell manipulation and light sheet-based 3D imaging. Histopathology

The facility offers histopathological services and training opportunities to the IRB scientific community and external public and private research centers, applying their experience to provide personalized and cost-effective support.
