Bifunctional therapeutic shRNAs for drug resistance eradication. 2shRNA aims to deliver a novel RNA-based therapeutic approach with great potential to simultaneously knockdown two signalling pathways in cancer cells. The existing proof-of-principle was designed to fight HER2 positive breast tumours. The approach has involved bioinformatics studies, cycles of chemical refinement and biological evaluation in cell cultures of breast cancer tumours.

Market Data

Problem: Acquired resistance is a major limitation for the successful treatment of breast cancer by targeted therapies. Several drug combinations for the simultaneous attack of primary and secondary pathways have been approved, but they present several problems such as cooperative toxicity effects, lack of efficacy and poor bioavailability, hampers their clinical use and forces the development of new combinatorial approaches. Solution: An alternative approach to traditional combined therapies consists in the simultaneous inactivation of primary and rescue pathways by using short RNA, which trigger the RNA interference machinery (both siRNAs and shRNAs), and avoid the expression of the genes of interest. If technical problems related mostly to biostability and delivery are solved, dual short RNA-based therapies may become the magic bullet to treat cancer.
